Abstract: Cancer evolution is characterized by extensive changes of regulatory landscapes, which are poorly understood at the functional level. Using genome-wide screening approaches based on transposon mutagenesis, regulatory elements of T cell acute lymphoblastic leukemia (T-ALL) were identified and the widespread roles of subtle gene regulation were functionally validated in cell lines and mice. Modelling human T-ALL heterogeneity in mice uncovered subtype-specific clonal structures, driver genes, pathway hierarchies and sequentialities.
